Nolvadex - non-steroid estrogen antagonist. Specifically binds to estrogen receptors in peripheral and tumor tissues. Thus, it eliminates the influence of estrogens produced in the body and stimulating the growth of cancer cells.

Slows the progression of the tumor process. After taking a single dose for several weeks, the ability to block estrogens will remain. Causes ovulation in women, in men with oligospermiaincreases the level of LH and FSH.

Pharmacokinetics

When taken orally, it is rapidly absorbed, and after 5-7 hours, the maximum concentration is created in the blood. 99% bound to blood proteins (albumin) Excretion occurs in two phases: the initial T 1/2 from 8 to 14 hours, the final T 1/2 - 7 days. It is excreted in feces, only a small part with urine.

Indications for use

Nolvadex is used for:

  • estrogen-dependent breast tumors in women during postmenopausal women;

Contraindications

Nolvadex tablets are contraindicated in:

  • hypersensitivity;
  • pregnancy
  • breastfeeding;
  • cataract;
  • pronounced thrombophlebitis;
  • leukopenia, thrombocytopenia.

Side effects

Nolvadex may cause the following adverse reactions:

Nolvadex tablets are taken orally without chewing, washed down with water. It is possible to take the entire dose at once in the morning or in two doses - in the morning and in the evening. At breast cancer 20-40 mg are prescribed, with endometrial cancer - 30-40 mg. The maximum diabetes is 40 mg. The treatment is carried out for a long time, often in combination with cytostaticand radiation therapy.

During the treatment period, blood coagulation, leukocytes, platelets, blood calcium level, and liver function are controlled. An ophthalmologist is examined quarterly and a gynecological examination is performed. The drug Nolvadex is ineffective in the presence of metastases(especially in the liver).

Overdose

It is manifested by an increase in side effects. Symptomatic treatment is carried out. The antidote does not exist.

Interaction

Nolvadex (Tamoxifen) enhances the effect anticoagulants. Concomitant use of the drug with estrogen-containing contraceptives causes a decrease in the action of both drugs.

Application with Tegafurcan lead to hepatitisand cirrhosis. Allopurinolalso enhances its hepatotoxic effect.

Joint reception with cytostaticssignificantly increases the risk of thrombosis.
Reception with thiazide diuretics increases the risk of hypercalcemia.

H2-histamine receptor blockers and antacidsmay cause premature dissolution of enteric tablets, so the time between taking these drugs should be 2 hours.
